Summary Telomeric fingerprinting was found to be highly differentiating for Paecilomyces fumosoroseus and Paecilomyces lilacinus isolates in comparison to intron splice site PCR and is therefore a good method for quality control of future products based
on these fungi. Although the telomeric restriction length polymorphisms correctly divided the isolates into their appropriate
species, further correlation with host range or geographical origin of the isolates was not found. In this respect, intron
splice site PCR was more informative taxonomically. The chromosome numbers inferred from telomeric fingerprints were seven

Abstract. Questions: How do physical microsite conditions of microsites affect germination and seedling survival in different successional stages? Do different species germinate in similar microsites in a given successional stage? Location: Coleman Glacier foreland, Mount Baker, Washington State, USA. Methods: Two methods were used to characterize safe sites. 1. Grids of 300 10 cm × 10 cm plots were located in four different age classes on the foreland. 2.105 pairs of plots, with and without seedlings of Abies amabilis, were located in each age class. For each plot we identified all seedlings and all individuals < 1 m tall. Microsite characteristics such as topography and presence of rocks or woody debris were noted for each plot. Microsite characteristics were compared between plots with and without each species. In addition we examined the effect of distance from seed sources on the presence of Alnus viridis seeds and seedlings in a newly disturbed area. Results: In early successional sites, seedlings of several species were positively associated with depressions and presence of rocks, and negatively associated with ridges. Patterns were generally consistent among species. In later succession, seedlings were not significantly associated with any microsite characteristics. For Alnus viridis, seed density decreased with distance from seed sources but seedling density did not. Conclusions: Because of harsh conditions in early succession, physical microsites are important, and most species have similar microsite requirements. In later succession, physical microsites characteristics are not as important and are more variable. Microsites appear to be more important than seed rain in controlling the distribution of Alnus viridis in early succession. 相似文献

The distributions of eight out of nine common species of waders (Charadrii) overwintering on UK estuaries have changed in association with recent climate change. These birds represent a high proportion of various populations from breeding grounds as far apart as Greenland to the west to high‐arctic Russia to the east. During warmer winters, smaller proportions of seven species wintered in south‐west Britain. The distributions of the smaller species show the greatest temperature dependence. The opposite was found for the largest species and no relationship was found for a particularly site‐faithful species. In north‐west Europe, the winter isotherms have a broadly north to south alignment, with the east being colder than the west. The average minimum winter temperatures across the UK having increased by about 1.5°C since the mid‐1980s, the temperatures on the east coast during recent winters have been similar to those of the west coast during the mid‐1980s. On average, estuaries on the east and south coasts of Britain have muddier sediments than those on the west coast and thus support a higher biomass of the invertebrate prey of waders. We suggest that, with global climatic change, the advantage gained by waders wintering in the milder west to avoid cold weather‐induced mortality is diminished. Consequently, more choose to winter in the east and thus benefit from better foraging opportunities. The implications of these results are considered in terms of a site‐based approach to wildlife protection used in Europe and elsewhere. 相似文献

The vertical distribution of dominant genera of bacterivorous nematodes to 150-cm depth in an aquic brown soil was compared after 14 years of four contrasting land uses, i.e., cropland-rice (CR), cropland-maize (CM), abandoned cropland (AC), and woodland (WL). The study was conducted at the Shenyang Experimental Station of Ecology, a Chinese Ecosystem Research Network (CERN) site in Northeast China. Data were analyzed using two-way analysis of variance with land use and depth as independent variables. More than 70% of Chiloplacus, Eucephalobus, and Monhystera spp. were present in the uppermost soil layer (0 to 5 cm) in the CR treatment. In contrast, Chiloplacus and Prismatolaimus spp. were distributed down to 100-cm depth in the AC and CM treatments, respectively. Differences in numbers of Acrobeles, Acrobeloides, Cephalobus, Chiloplacus, Eucephalobus, Monhystera, Plectus, and Prismatolaimus were found among land uses and at various depths. Soil C and N were correlated positively with numbers of Monhystera and Plectus in the CR treatment, Acrobeloides in the CM treatment, and Acrobeles and Acrobeloides in the AC treatment. Soil pH was correlated negatively with Monhystera, Plectus (CR), and Acrobeloides (CM, AC). The relationship of pH with Acrobeles depended on land use: positive in the WL treatment and negative in the AC treatment. Our results suggested that Cephalobus and Prismatolaimus in the CR treatment, and Chiloplacus and Prismatolaimus in the WL treatment, were insensitive to soil properties measured. Differences in vertical distribution should be considered when studying dominant bacterivorous nematode genera among land uses. 相似文献

转录起始位点的计算定位是基因转录调控研究的重要内容,但现有方法的识别性能较低。文章作者在已有原核启动子识别算法的基础上,提出了一种基于滑动窗口的原核转录起始位点计算定位方法,通过在合理限定的定位范围内对序列进行滑动扫描,来预测转录起始位点的位置。首先根据窗口序列的交迭组分特征和启动子其它特征分别建立二次判别分类器,用其计算对应位置的似然得分,再利用转录起始位点与翻译起始位点的间隔经验分布信息对似然得分进行修正,最后依照似然得分的分布情况由阈值定位算法确定预测位置。对大肠杆菌真实序列数据的测试结果表明,该定位算法可实现对真实转录起始位点位置的有效预测,与已有算法相比,当敏感性指标同为0.85左右时,特异性指标可从0.20提高至0.65,从而使得定位准确率提高了约20个百分点。 相似文献

The spawning site of the fluvial eight-barbel loach, Lefua sp. (sensu Hosoya, 1993; Japanese name: nagare-hotoke-dojo), an endangered species, was investigated by searching for fertilized
eggs in June and July 1995–1997 in an upper reach of the Kako River, Hyogo Prefecture, Japan. We obtained one egg each by
a random sampling with agitating the river bottom (80 trials) and by collecting drift samples (89 trials). This finding indicates
that the eggs were not scattered on the bottom or into the water column after being spawned. Two individual males were observed
to patrol around particular interstices occurring under the buried cobbles and under the boulders that formed the riverbank.
Males pecked other individuals that approached around the interstices. We obtained 9 and 15 eggs, respectively, from two such
interstices in only two collection trials. The interstices were suggested to function as the spawning sites of this endangered

-Amylases are endo-acting retaining enzymes of glycoside hydrolase family 13 with a catalytic (β/)8-domain containing an inserted loop referred to as domain B and a C-terminal anti-parallel β-sheet termed domain C. New insights integrate the roles of Ca2 + , different substrates, and proteinaceous inhibitors for -amylases. Isozyme specific effects of Ca2 + on the 80% sequence identical barley -amylases AMY1 and AMY2 are not obvious from the two crystal structures, containing three superimposable Ca2 + with identical ligands. A fully hydrated fourth Ca2 + at the interface of the AMY2/barley -amylase/subtilisin inhibitor (BASI) complex interacts with catalytic groups in AMY2, and Ca2 + occupies an identical position in AMY1 with thiomaltotetraose bound at two surface sites. EDTA-treatment, DSC, and activity assays indicate that AMY1 has the highest affinity for Ca2 + . Subsite mapping has revealed that AMY1 has ten functional subsites which can be modified by means protein engineering to modulate the substrate specificity. Other mutational analyses show that surface carbohydrate binding sites are critical for interaction with polysaccharides. The conserved Tyr380 in the newly discovered 'sugar tongs' site in domain C of AMY1 is thus critical for binding to starch granules. Furthermore, mutations of binding sites mostly reduced the degree of multiple attack in amylose hydrolysis. AMY1 has higher substrate affinity than AMY2, but isozyme chimeras with AMY2 domain C and other regions from AMY1 have higher substrate affinity than both parent isozymes. The latest revelations addressing various structural and functional aspects that govern the mode of action of barley -amylases are reported in this review. 相似文献
